Medicinal Chemistry Core Facility (Oregon Health and Science Unversity ) at Medicinal Chemistry Core, we help OHSU researchers investigate interactions between small molecules and biological systems. To accomplish this, Med Chem Core provides medicinal chemistry, chemical biology and computational expertise to facilitate biologically oriented innovation.
We have several on-going projects that relate to TB, Malaria, Antiviral, Cardiovascular and Macular degeneration.
Taking TB project as an example, Tuberculosis (TB) is one of the major causes of morbidity and mortality worldwide for centuries. Control of the spread of Mycobacterium tuberculosis infection remains a priority to public health, in particular, to those in the armed forces who are deployed to places with high TB infections.
Recent research found that MAIT cells, mucosal associated invariant CD8+ T-cells subsets, enriched in airways, are uniquely activated by vitamin B metabolites presented by MR1, major histocompatibility class I-like antigen-presenting molecule, consequently, initiating IFN-γ production ex vivo in response to cells infected with Mycobacterium tuberculosis. Small molecules, other than vitamin B metabolites could potentially bind to MR1 based on the analysis of its binding mode and binding space. We hypothesized that small molecules, other than vitamin B metabolites which are riboflavin and folic acid pathways derivatives, could activate MAIT cells presented by TB selective MR1, thus provide the basis for a new TB vaccine strategy.
Our preliminary data demonstrated that MR1 can present structurally diverse small molecules. Using mass spectrometry analysis of recombinant MR1 loaded with ligands in the context of bacterial infection, we have detected several novel small molecules as MAIT cell activating ligands.
In this project, you are expected to design and synthesis the novel small molecules as MAIT cell activating ligands.
1. Relevant academic background and Laboratory experience in Organic Chemistry, Medicinal Chemistry or other related discipline; Master or Ph.D candidate, Ph.D is preferred.
2. Financial supports from the Chinese government that cover his/her living expenses in USA. Preference will be given to scholars who have research experience on organic chemistry and publications.
3. The candidate must show demonstrated capability to work within a highly collaborative environment.
